Using micropatterns of BMP-2 and fibronectin to recreate the stem cell niche

Abstract

In vivo, stem cells reside within a niche microenvironment which regulates at a highly localized level both self-renewal and differentiation. This occurs through cell-cell interactions, the action of growth factors, and the properties of the extracellular matrix. Recreating the stem cell niche in vitro is a crucial aspect in understanding how the microenvironment controls stem cell behavior. Here we used microcontact printing to generate micropatterns of growth factors (BMP-2) and extracellular matrix proteins (Fibronectin) on a soft biomimetic films, and thus recreate a simplified niche model to study the factors influencing osteoblastic differentiation.
