Fabrication of cellular micropatterns using a functional substrate with convertible cell-adhesive surface properties

Abstract

Positioning of cells in a desired pattern on a substrate is an important technique for cell-based technologies including the fundamental investigation of cell functions, tissue-engineering applications, and the fabrication of cell-based biosensors and cell arrays.Although several successful strategies for creating cellular micropatterns on substrates have been demonstrated, these methods require the special and expensive equipment or materials and cannot apply to nonadherent cells. We established a novel facile fabrication method for cellular micropatternsincluding the nonadherent cells by using serum albumin which is the most abundant protein in blood plasma.We prepared a water-insoluble, cross-linked albumin film possessing the properties of native albumin, such as resistance to cell adhesion and drug binding ability. Then, we found that the cell non-adhesive surface property could be changed easily to allow cell adhesion by exposing the cross-linked albumin film to UV light or cationic polymer solution.Cellular micropatterns were created on substrate by utilizing this convertible cell-adhesion property of albumin film. In addition, micropatterns of the nonadherent cellswere also successfully created by combining the albumin film and cell membrane anchoring reagent which can interact with membranes of living cells without cytotoxicity.Our technique is promising for the creation of the complex cellular micropattern composed of multiple types of cells including nonadherent cells.
